چکیده

Activation of C3 by nylon 6.6 and polymethyl methacrylate (PMMA) polymers in whole canine serum was investigated using Laurell electroimmunoassay and antigen-antibody crossed immunoelectrophoresis (XAA). Canine sera were utilized because most artificial blood contact materials used in prosthetic devices are tested in dogs. The measurements were made possible by converting the polymers to microparticles, increasing the available surface area to approximately 200,000 sq cm/g. Both polymers activated C3 within a few minutes of serum contact at 39’C. the canine biologic temperature, after which no further development was seen. Nylon activation, studied in detail, increased with increasing surface area up to a saturation level of about 20% conversion reached at approximately 350 sq cm/mI polymer surface/serum

Absence of conformational change in complement factor 3 and factor XII adsorbed to acrylate polymers is related to a high degree of polymer backbone flexibility.

In previous investigations, the authors have examined the adsorption of albumin, immunoglobulin, and fibrinogen to a series of acrylate polymers with different backbone and side-group flexibility. The authors showed that protein adsorption to acrylates with high flexibility, such as poly(lauryl methacrylate) (PLMA), tends to preserve native conformation. In the present study, the authors have c...

C3 adsorbed to a polymer surface can form an initiating alternative pathway convertase.

Contact between blood and a biomaterial surface induces an immediate complement-mediated inflammatory response. Under these conditions, the alternative pathway of complement is often initiated and amplified on the biomaterial surface. Adsorption of a protein such as C3 to a polymer surface induces conformational changes in the protein. Based on the expression on adsorbed C3 of conformational ne...

Characterizing films of polyethylene blends: An application of colorimetric parameters measurements

This study reports an application of instrumental color measurement for quantifying and comparing the optical properties of high density polyethylene HDPE and low density polyethylene LDPE blend films and the effect of blending ratio and miscibility on these properties. The technique of spectrophotometry in transmission and reflectance modes can be used as a versatile tool to evaluate the color...

On the binding of complement to solid artificial surfaces in vitro.

Since the realization of a complement activation capacity by artificial surfaces upon contact with blood, a common belief has evolved that charged nucleophilic surface groups such as amine (-NH2) and hydroxyl (-OH) react with and eventually bind to the internal thioester in complement factor 3 (C3). A covalent amide or ester linkage is thereby supposed to form between C3b and the surface itself...
